Abstract
Insect cell lines are frequently
used for expression of
recombinant glycoproteins,
including vaccines. Using two
independent approaches, we
show that the endogenous Nglycomes of High Five and Sf9
cells contain species-specific
anionic N-glycan structures with
different combinations of
phosphorylcholine, fucose,
hexuronic acid and/or pentose.
Glycans with phosphorylcholine
and fucose epitopes were also
found on influenza hemagglutinin
and SARS-CoV-2 Spike
expressed in either cell line. Our
data highlight the need for open
search strategies for identifying
glycans present on recombinant
glycoproteins from nonmammalian sources.
